Stem Cells Restore Cognitive Abilities Impaired By Brain Cancer ...

Human neural stem cells are capable of helping people regain learning and memory abilities lost due to radiation treatment for brain tumors, a UC Irvine study suggests.

Research with rats found that stem cells transplanted two days after cranial irradiation restored cognitive function, as measured in one- and four-month assessments. In contrast, irradiated rats not treated with stem cells showed no cognitive improvement.

?Our findings provide solid evidence that such cells can be used to reverse radiation-induced damage of healthy tissue in the brain,? said Charles Limoli, a UCI radiation oncology professor.

Study results appear in the July 15 issue of Cancer Research, a journal of the American Association for Cancer Research.

Radiotherapy for brain tumors is limited by how well the surrounding tissue tolerates it. Patients receiving radiation at effective levels suffer varying degrees of learning and memory loss that can adversely affect their quality of life.

?In almost every instance, people experience severe cognitive impairment that?s progressive and debilitating,? Limoli said. ?Pediatric cancer patients can experience a drop of up to three IQ points per year.?

For the UCI study, multipotent human neural stem cells were transplanted into the brains of rats that had undergone radiation treatment. They migrated throughout the hippocampus ? a region known for the growth of new neurons ? and developed into brain cells.

Researchers assessed the rats one month and four months after transplantation, noting enhanced learning and memory abilities at both intervals.

Additionally, they found that transplanting as few as 100,000 human neural stem cells was sufficient to improve cognition after cranial irradiation. Of cells surviving the process, about 15 percent turned into new neurons, while another 45 percent became astrocytes and oligodendrocytes ? cells that support cerebral neurons.

Most notably, Limoli said, he and his colleagues discovered that about 11 percent of the engrafted cells expressed a behaviorally induced marker of learning, indicating the functional integration of those cells into memory circuits in the hippocampus.

?This research suggests that stem cell therapies may one day be implemented in the clinic to provide relief to patients suffering from cognitive impairments incurred as a result of their cancer treatments,? Limoli said. ?While much work remains, a clinical trial analyzing the safety of such approaches may be possible within a few years, most likely with patients afflicted with glioblastoma multiforme, a particularly aggressive and deadly form of brain cancer.?

Munjal Acharya, Lori-Ann Christie, Mary Lan and Erich Giedzinski of UCI and John Fike and Susanna Rosi of UC San Francisco contributed to the study, which was funded by the California Institute for Regenerative Medicine, the National Institutes of Health and the U.S. Department of Energy.

'Harry Potter' Encyclopedia In The Works

J.K. Rowling announces she will donate all royalties to charity.

Last week, fans of J.K. Rowling were treated to the title and release date of the "Harry Potter" author's new novel, "The Casual Vacancy." This week is sure to have fans buzzing again as it brings the news that she's started working on a "Harry Potter" encyclopedia.

Rowling answered a fan question regarding the highly anticipated Potter lexicon on the "frequently asked questions and rumours" page of her website.

"For a long time I have been promising an encyclopaedia of Harry's world, and I have started work on this now — some of it forms the new content in Pottermore. It is likely to be a time-consuming job, but when finished I shall donate all royalties to charity."

Back in 2008, Rowling and Warner Brothers won a lawsuit against Steve Vander Ark, the publisher of the Harry Potter Lexicon, who attempted to release a book along the same lines.

Rowling also addressed the biggest burning fan question of all on her website: Will there ever be another "Harry Potter" novel?

"I have always refused to say 'never' to this question," the British author wrote, "because I think it would be foolish to rule out something I might want to do in a few years' time. However, I have no immediate plans to write another Harry Potter novel, and I do think that I have rounded off Harry's story in seven published books."

With the sheer volume of a project like the Harry Potter encyclopedia, it's unlikely it will be available anytime soon. But Rowling's first adult novel, "The Casual Vacancy," is set to be released on September 27, 2012. The novel follows a quaint town in the wake of a death that leaves an empty seat on the parish council. Despite appearances, the town isn't all it seems. Behind closed doors it's in a continual state of war and this sudden death leaves it in its biggest war yet.

Should J.K. Rowling write another "Harry Potter" book? Leave your comment below!?

New insights into when beach sand may become unsafe for digging and other contact

ScienceDaily (Apr. 11, 2012) ? With summer days at the beach on the minds of millions of winter-weary people, a new study provides health departments with information needed to determine when levels of disease-causing bacteria in beach sand could pose a risk to children and others who dig or play in the sand. The report appears in ACS' journal Environmental Science & Technology.

Tomoyuki Shibata and Helena M. Solo-Gabriele explain that disease-causing bacteria from sewage can cause skin infections and gastrointestinal (GI) disorders in people who come into contact with contaminated water. The U.S. Environmental Protection Agency (EPA) has guidelines to determine when microbe levels in water are high enough to pose an unacceptable risk of GI illness for contact with both ocean water and freshwater. Microbes, however, tend to concentrate in higher levels in beach sand -- to the point where one previous study found that the sand on one fingertip, placed in the mouth, had enough germs to cause GI illness. No guidelines exist to determine when contact with beach sand might be too risky for children and others who play in beach sand, digging in it or being buried in the sand. The scientists set out to fill that knowledge gap.

The scientists used millions of computer simulations and measurements of disease-causing microbes at beaches in California and Florida to determine how many bacteria would have to be present in beach sand to exceed the EPA's guideline for water. In doing so, they established "reference levels" for beach sand that correspond to the EPA risk guidelines for water. The focus of children at the beach environment is especially important, due to play behavior at beach sites that would increase a child's exposure, the scientists noted.

The authors acknowledge funding from the National Science Foundation and the National Institute of Environmental Health Sciences.

Prosecutors face hurdles in Trayvon Martin case

SANFORD, Fla. (AP) ? After an extraordinary public campaign to make an arrest in the shooting of an unarmed black teen, a Florida prosecutor came back with a murder charge in the case that has galvanized the nation for weeks.

But prosecutors face steep hurdles to win a second-degree murder conviction against neighborhood watch volunteer George Zimmerman in the killing of Trayvon Martin, experts say. They will have to prove Zimmerman intentionally went after Martin instead of shooting him in self-defense, refute arguments that a Florida law empowered him to use deadly force and get past a judge's ruling at a pretrial hearing.

Zimmerman, 28, who turned himself in at a county jail Wednesday after prosecutor Angela Corey announced the charge, was to appear before a magistrate Thursday and plead not guilty in the Feb. 26 shooting of the 17-year-old that set off a nationwide debate about racial profiling and the rights to self-defense.

"He is concerned about getting a fair trial and a fair presentation," his attorney, Mark O'Mara said. "He is a client who has a lot of hatred focused on him. I'm hoping the hatred settles down ... he has the right to his own safety and the case being tried before a judge and jury."

Legal experts said Corey chose a tough route with the murder charge, which could send Zimmerman to prison for life if he's convicted, over manslaughter, which usually carries 15-year prison terms and covers reckless or negligent killings.

The prosecutors must prove Zimmerman's shooting of Martin was rooted in hatred or ill will and counter his claims that he shot Martin to protect himself while patrolling his gated community in the Orlando suburb of Sanford. Zimmerman's lawyers would only have to prove by a preponderance of evidence ? a relatively low legal standard ? that he acted in self-defense at a pretrial hearing to prevent the case from going to trial.

There's a "high likelihood it could be dismissed by the judge even before the jury gets to hear the case," Florida defense attorney Richard Hornsby said.

Corey announced the charges Wednesday after an extraordinary 45-day campaign for Zimmerman's arrest, led by Martin's parents and civil rights activists, including the Rev. Al Sharpton and the Rev. Jesse Jackson. Protesters wore hooded sweatshirts like the one Martin had on the night of the shooting. The debate reached all the way to the White House, where President Barack Obama observed last month: "If I had a son, he'd look like Trayvon."

Corey would not discuss how she reconciled conflicting accounts of the shooting by Zimmerman, witnesses and phone recordings that indicated Martin thought Zimmerman was following him.

"We do not prosecute by public pressure or by petition. We prosecute based on the facts on any given case as well as the laws of the state of Florida," Corey said.

Martin's parents expressed relief over the decision to prosecute the person who shot their son.

"The question I would really like to ask him is, if he could look into Trayvon's eyes and see how innocent he was, would he have then pulled the trigger? Or would he have just let him go on home?" said his father, Tracy Martin.

Many attorneys said they had expected the prosecutor to opt for the lesser charge of manslaughter. The most severe homicide charge, first-degree murder, is subject to the death penalty in Florida and requires premeditation ? something all sides agreed was not present in this case.

"I predicted manslaughter, so I'm a little surprised," said Michael Seigel, a former federal prosecutor who now teaches law at the University of Florida. "But she has more facts than I do."

O'Mara, Zimmerman's attorney, said his client would plead not guilty and invoke Florida's so-called "stand your ground" law, which gives people wide latitude to use deadly force rather than retreat during a fight.

The confrontation took place in a gated community where Martin was staying with his father and his father's fianc?e. Martin was walking back in the rain from a convenience store when Zimmerman spotted him and called 911. He followed the teenager despite being told not to by a police dispatcher and the two got into a struggle.

Zimmerman told police Martin punched him in the nose, knocking him down, and then began banging the volunteer's head on the sidewalk. Zimmerman said he shot Martin in fear for his life. Sanford police took Zimmerman, whose father is white and whose mother is Hispanic, into custody the night of the shooting but released him without charging him.

A judge could dismiss the charge based on the "stand your ground" law, legal experts said. But some experts say the judge will also be under tremendous pressure to let the case go forward.

"Judges are not likely to take that out of the hands of the jury," said Florida defense attorney Randy Reep.

Other attorneys weren't surprised that Corey went for the maximum.

"Prosecutors look for leverage. They'll typically overcharge knowing that gives them wiggle room for plea discussions," said Derek Byrd, incoming president of the Florida Association of Criminal Defense Lawyers. "She knows that she could offer him manslaughter at some point or get in front of a jury that could split the verdict and agree on a lesser offense."

Corey wouldn't discuss how she arrived at the charges or disclose other details of her investigation. The prosecutor in Jacksonville was appointed to handle the case by Republican Gov. Rick Scott after the local prosecutor disqualified himself.

The U.S. Justice Department's civil rights division is conducting its own civil rights investigation.

Tensions had risen in Sanford, a town of 50,000 outside Orlando. Someone shot up an unoccupied police car Tuesday outside the neighborhood where Martin was killed. Outside of Sanford City Hall on Wednesday, Stacy Davis, who is black, said the arrest "is not a black or white thing for me. It's a right or wrong thing."

In Washington, Martin's family pleaded for calm in response to the decision. But Martin's mother, Sybrina Fulton, clasped hands and smiled in relief when she heard Corey utter the words "second-degree murder" on television.

"We wanted an arrest and we got it," Fulton said later. "Thank you Lord, Thank you, Jesus."

Do Mobile Phones Cause Cancer?

The World Health Organization announced yesterday that our beloved mobile communication devices may cause cancer, which puts mobile (cell) phones in the same category as car exhaust, chloroform, and lead. This news comes after studies showing that exposure to a transmitting phone pressed against your ear for 50 minutes causes increased brain glucose and activity in the areas closest to the device. The form of radiation emitted from these devices is referred to as non-ionizing, which is similar to that of a low-powered microwave oven as opposed to an X-ray. Obama-Romney showdown starts with a harsh tone

MENDENHALL, Pa. (AP) ? The 2012 presidential general election has begun. It won't be pretty.

Tuesday marked Day One, in essence, of the contest between the two virtually certain nominees, Republican Mitt Romney and President Barack Obama. Rick Santorum's departure removed the last meaningful bump from Romney's path to the GOP nomination. Romney and Obama wasted no time in portraying the voters' choice in dire, sometimes starkly personal terms.

With Obama saddled with a still-ailing economy and a divisive health care law, and Romney riding a wave of blistering TV ads, the fall election is unlikely to dwell on "hope," ''change" and other uplifting themes from four years ago. Much of the nation's ire then was aimed at departing President George W. Bush, and Obama had no extensive record to defend.

The landscape is much different now. Americans face nearly seven months of hard-hitting jabs and counterpunches between the two parties' standard-bearers.

Romney, the former Massachusetts governor making his second presidential bid, attacked Obama with gusto Tuesday in his two public events that followed Santorum's surprising announcement.

Campaigning in Pennsylvania, where an April 24 GOP primary is suddenly less important than its likely role as a battleground state this fall, Romney portrayed Obama as a weak leader who apologizes for America's greatness and prefers European-style socialism over robust free enterprise. Obama's allies call such claims nonsense.

"The right course for America is not to divide America," Romney told a GOP dinner gathering in Mendenhall, near Philadelphia. "That's what he's doing," he said of Obama. "His campaign is all about finding Americans to blame and attack, and find someone to tax more, someone who isn't giving, isn't paying their fair share."

He said Obama prefers "a government-centered society."

Obama, campaigning in Florida, said the choice this fall will be as stark as in the 1964 contest between Lyndon Johnson and Barry Goldwater, which resulted in one of the biggest Democratic landslides ever. That election included dramatic and controversial moments, such as Goldwater's defense of "extremism in the defense of liberty" and a devastating TV ad suggesting a Goldwater presidency would lead to nuclear war.

Obama didn't mention Romney by name. His top aides have shown less restraint, however.

Obama campaign manager Jim Messina said in a statement after Santorum's withdrawal: "It's no surprise that Mitt Romney finally was able to grind down his opponents under an avalanche of negative ads. But neither he nor his special interest allies will be able to buy the presidency with their negative attacks. The more the American people see of Mitt Romney, the less they like him."

Other Obama campaign officials have mocked Romney's wealth and called him out of touch with average Americans.

Romney and his allies, including a potent super PAC, have proved their ability to raise millions of dollars to air brutally effective attack ads, which crippled Santorum and Newt Gingrich in the GOP primary contests. Obama will raise many millions, too, and few doubt that he will hit Romney hard.

On Tuesday, Romney made clear that he will go after Obama's character as well as his record. In speeches in Mendenhall and Wilmington, Del., Romney said Obama isn't merely inept at economic policy, he actively dislikes business.

Obama "is clearly trying to hide from us what he intends to do," Romney said in Wilmington. "He's going to hide. And it's my job to seek."

Romney made similar remarks last month. Now, with Santorum off the stage and Gingrich and Ron Paul hardly a factor, there are no intra-party distractions to dilute such comments. Romney and Obama are fully engaged, one-on-one, at a much earlier stage than in 2008, when Obama had to parry Democratic rival Hillary Rodham Clinton throughout the summer before fully turning to Republican John McCain.

Even then, Bush's unpopularity helped fuel Obama's campaign and deflected some of the anti-GOP sentiment away from the actual nominee.

This time, the incumbent president is on the ballot, with unemployment above 8 percent. The tea party, which didn't exist in 2008, is a potent and unpredictable force.

And Romney suddenly is free of meaningful primary worries. That leaves him able to focus the full force of his fundraising and campaigning skills against Obama.

Should You Consider Medical Bankruptcy? - Loan Insurance ...

From a lawful standpoint, there is no provision particularly for medical bankruptcy as the court treats all debt scenarios in similar ways. Even so, you could testify that your accumulated debt was mainly caused by medical expenditures if you desire to organize for bankruptcy.

Anyone of wisdom can easily realize that simply one illness that puts you into the hospital can dangerously drain anyone?s financial situation and this is why numerous folks from all walks of life are strained to sell their assets or spend all their wages on clinic charges and the like. According to a examine done a couple of years ago, more than fifty percent of all the bankruptcy petitions filed in federal court are connected to medical past due debt and hounding creditor threats to pay medical bills.

Offered the tide state the financial system is in, it?s safe and sound to say that the scenarios of medical bankruptcy have increased considering that then. If you?re in the identical situation, you ought to know that filing for bankruptcy is not usually the greatest alternative to go with.

First of all, applying for Chapter 7bankruptcy is a lengthy course of action (usually long lasting several months) and wiping your debt under this scheme involves the insolvency of your non-essential assets. If you?re wondering what that means, the appointed trustee assigned to your case will set up every single thing for sale (except your house and car) as a way to decide your debt.

Furthermore, the credit scorers could very well say no to the filing of medical bankruptcy and push for your case to be dismissed by the court leaving you high and dry without a prayer. In the event that your effort for a Chapter 7 Bankruptcy request does not push through, the next step is to step on to a Chapter 13 Bankruptcy set up.

You?re also ready to be positioned for a dangerous state of affairs with this selection because you have no control over the terms under a Chapter 13 bankruptcy process. Even though you won?t be liquidating your assets and estate under this provision, you will be put in a 3-5 year payment plan which is by and large awfully tough to adhere to.

You don?t get to indicate how much you want to pay every single month nor do you have any influence about the length of the full plan. What?s extra difficult is that you still have to to pay up even if you get sick or injured.

As such, taking on creditors in court through the best of all bankruptcy courses of action is a risky endeavor with various unknown factors that could go incorrect to your finances and be extremely regretful and damaging for the rest of your life. Even if you successfully manage to file medical bankruptcy, there are long-term penalties such as a broken credit score rating for over a 10-year period as well as the disgrace of bankruptcy looming over your head for the rest of your entire life.

As such, you can not rely on a satisfactory bankruptcy as the sole means of settling your past due bills. Although having an attorney is crucial, he or she may well not be aware (or be inclined to recommend) other avenues of service aside from the bankruptcy services they offer.

It?s significant to explore other alternatives aside from filing for a decent medical bankruptcy ? you may perhaps be overlooking other imperative assets which may perhaps give you further force in opposition to the creditors who are distressing to you.

There are in actuality, third-party consultation programs online which can give you extra assistance on top of the lawyer?s services. With their advantage, you can shorten the course of action and save money on official fees in the long run by learning about the debt resolution course of action in general (and not just bankruptcy).

Consider, you can?t afford not to see the big picture. creating an effort to educate oneself is empowering and will help you take the greatest course of action feasible.

Are you wanting to File Bankruptcy Yourself and get a Medical Bankruptcy plan? Make sure you check Vernon Mortensens? website for info you must know to prevail.

The primary aim of such firms is to keep an eye on e-mail services, credit card processing and operational monitoring. Web sites must operate throughout the year without any complications.

One vital role played by web hosts is supplying a reliable technical support system. Web sites can require maintenance during off-peak hours. For this reason, it is very important for a web host to provide web site upkeep even during non-working hours. There is an enormous worldwide demand for web host companies. In fact, a new ?ecommerce website? is launched daily.

Web hosts assist in smooth functioning of shopping carts. These days, people prefer paying for goods and services through their credit cards. To ensure that the payments made through credit cards are safe, web hosts provide the required support. The field of electronic commerce is growing like never before.

It is important to understand ecommerce hosting before delving into why web hosting is so important. Some companies sell web server space and services to other companies, who in turn use the services to sell products and make money online. These base companies are called web hosts and web hosting firms. Ecommerce shopping cart software is also available from web hosting companies.

Web hosts provide templates for creating online catalogs and they gather details of financial transactions between businesses. ?Ecommerce hosting? firms can help you increase your profits. Finding a good ?ecommerce hosting? company is not difficult to do. The Internet has a plethora of information about firms that do ?ecommerce hosting.?

Before choosing a particular web hosting company, you must consider important factors like security, features and of course your budget. You can also search for the addresses and contact numbers of ?web hosting companies? in local newspapers and magazines. Your friends and relatives (who have knowledge of web hosting) can also help you find the best ?web hosting company? in your town or city.

New ecommerce websites are created each and every day. Internet shopping and electronic commerce is a rapidly growing area of business. Since people like to use credit cards when shopping this way it is important to have great running shopping carts.

Autism, Standardized Testing, and Hey, Why Don't We All Just Move ...

There?s a new thing I?m doing that forces the teachers to remember that I?m part of Connor?s ?team? at school, and it keeps me informed about any issues that are going on. What I do is I meet with his resource teacher every month now, and it seems to be working.

Last week was our monthly meeting, and we discussed progress and challenges. Towards the end, we somehow got on the subject of testing, and the standardized testing that starts in third grade. I casually mentioned that Connor didn?t need the extra pressure, and that I would be opting him out of testing when the time came.

?But you can?t opt-out of state testing?, she said.

?Yes I can. People do it. There is a way to opt-out.?

And the kicker was when she said, ?I?ve never heard of anyone in this district opting out of testing.?

Now granted, she teaches K-2nd, and is not directly involved with testing, so she probably isn?t up-to-speed on things. And she did agree with me about how standardized testing has had a negative impact on teaching.

But damn. A teacher wasn?t even aware that you could not be forced to take the standardized tests.

Not that opting-out is right for everyone. But I happen to believe that standardized testing has narrowed the scope of teaching so that teachers are focusing more on test-specific material, and losing out on other teaching opportunities. It forces them to keep a pretty rigid pace in class, which means that they can?t afford to spend an extra day of class time exploring Mark Twain?s other literary contributions if a class has shown particular interest in him after reading The Adventures of Tom Sawyer.

In addition, many school districts instruct their teachers to identify their ?bubble kids?, children on the cusp of meeting proficiency but needing extra help, and putting in extra time and focus on those children. This leaves children that are further behind and desperately in need of extra help left behind.

You know what else is interesting about standardized testing? There?s about four companies that produce the bulk of the testing materials, and they are making an ass-ton of money. Which means, lots. Lots of money being made to produce the testing materials mandated by the government under No Child Left Behind. Money coming out of our school districts. Money that isn?t being used to provide services for special needs kids, or extra support for typical children that need additional help.

In addition, these tests aren?t necessarily making our kids any smarter. Finland continues to score top marks in education, year after year. They do not have standardized testing, but instead assign less homework and promote more creative play. They also have no private schools, even on the college level. So people attend school equally from elementary to college. The most interesting quote I read was that ?education has been seen first and foremost not as a way to produce star performers, but as an instrument to even out social inequality.? But see, that?s not what we?re about here in America. We?re about individual achievement and standing out.

But I can put all that to the side when I consider standardized testing for Connor. The thing is, he?s already stressed from the pressures to keep up with his peers. He?s always just a bit behind the others, and works hard to keep it together at school all day, only to come home and do more homework at night, in an effort to ?catch up?. He has learned so much this year, and has come so far, but it has not been easy. When I think about adding the intense pressure of testing to that load, I can?t even fathom how he would manage.

And why should he? He is learning, he is working as hard as he is able to work. That is enough for me.

In a year-and-a-half, I might be the first and only parent in our school district to opt their child out of testing. Am I worried?? Nah, I?ve been raising a child with autism for almost 7 years now, they don?t scare me. But it is Texas, so if you don?t hear from me?.

Here are some official references and stuff, because I?m a sophisticated writer like that.

Goal Setting Kick-Start Guide | Self Improvement Blogs from ...

There are 5 essential, but simple actions to follow, which I believe are necessary for seemingly impossible objectives to be achieved.

Step One

Devote some quiet time and write down exactly what the change or alterations will probably be and why you would like them, this is the initial essential step towards achieving goals. At this stage, write only what it really is you?d like to obtain and why. ?The ?how?s? and ?when?s? can come later.

Step Two

This step too is quite essential, and it has almost everything to complete with you accessing a state of inner peace. The peace I write about has small to do with listening to relaxation music, important oils inside a burner or harps playing (though they can assist). Achievement of your objective needs getting peace via a quiet mind and inner calmness regardless of the chaos about you and how lengthy this chaos reigns.

Whenever you have this (peace) you?ll have higher clarity, your inventive self will make itself known, you?ll gain foresight into how you may achieve your outcome, concentrate, and negativity will have minimal impact.

Step Three

This important step is about gifting oneself with a fresh outlook on life. Taking a look at items within a completely different way and from a brand new angle will give you a a lot more international perspective revealing greater possibilities.

Step Four

By now you realize just what you want; you can step into profound inner peace at will; have picked up wonderful self-help and self-healing tools; eliminated limiting beliefs; and worked out your values. ?Now it is time to finally pinpoint specifically how you are going to go about achieving your purpose.

Step Five

This final but crucial step (they?re all crucial) will be the calculation of when your outcome will lastly eventuate. ?If you?ve followed the prior actions adding Step Number 4 into the equation, you?ll be able to establish the time you?ll be able to celebrate in a really, large way of finally reaching the pot in the end of the rainbow!

Any person can attain objectives by means of inner peace, perseverance, patience, flexibility and often viewing circumstances from an completely different angle. If you would like to read my 100 Day Challenge Review, head over to www.100daychallengereview.com.

Myanmar's Suu Kyi reported winning historic vote

YANGON, Myanmar (AP) ? She struggled for a free Myanmar for a quarter-century, much of it spent locked away under house arrest. Now, the Nobel Peace Prize laureate whose nonviolent campaign for democracy at home transformed her into a global icon is on the verge of ascending to public office for the first time.

Aung San Suu Kyi, 66, was elected to parliament Sunday in a historic victory buffeted by the jubilant cheers of supporters who hope her triumph will mark a major turning point in a nation still emerging from a ruthless era of military rule.

If confirmed, the election win will also mark an astonishing reversal of fortune for a woman who became one of the world's most prominent prisoners of conscience. When she was finally released in late 2010, just after a vote her party boycotted that was deemed neither free nor fair, few could have imagined she would make the leap from democracy advocate to elected official in less than 17 months, opening the way for a potential presidential run in 2015.

But Myanmar has changed dramatically over that time. The junta finally ceded power last year, and although many of its leaders merely swapped their military uniforms for civilian suits, they went on to stun even their staunchest critics by releasing political prisoners, signing cease-fires with rebels, relaxing press censorship and opening a direct dialogue with Suu Kyi ? whom they tried to silence for decades.

Results continued to trickle in early Monday from poll watchers within Suu Kyi's party, and spokesman Han Than said the opposition had won at least 43 of the 44 parliament seats it had contested. Those included four in the capital, Naypyitaw, considered a stronghold of the ruling party, whose leaders helped build the city.

The party's foray into electoral politics was its first since 1990 ? when it won a landslide that was promptly annulled by the army.

"It's the people's victory! We've taught them a lesson!" a shopkeeper who goes by the single name Thien said late Sunday, when it became clear the opposition was going to sweep the poll. He was among thousands watching outside the party's crumbling headquarters as a digital signboard repeatedly flashed news that Suu Kyi had won the Kawhmu constituency south of Yangon.

The crowds swelled as night fell, blocking traffic on the road. Some chanted "We won! We won!" Others clapped, danced, waved party flags and held their fingers aloft in V-for-victory signs. One official party message even told them not to gloat.

The results must be confirmed by the government's electoral commission, however, which has yet to release any outcome and may not make an official declaration for days.

U.S. Secretary of State Hillary Rodham Clinton congratulated Myanmar for holding the poll. Speaking at a news conference in Istanbul, Turkey, she said Washington was committed to supporting the Southeast Asian nation's reform effort.

"Even the most repressive regimes can reform, and even the most closed societies can open," she said.

The topdown revolution has left Myanmar befuddled and wondering how it happened ? or at least, why now? One theory says the military-backed regime had long been desperate for legitimacy and a lifting of Western sanctions, and its leadership had quietly recognized that their impoverished country, formerly known as Burma, had fallen far behind the rest of skyscraper-rich Asia.

Sunday's by-election was called to fill 45 vacant seats in Myanmar's 664-member bicameral assembly, and the military-backed government had little to lose by holding it. The last vote had already been engineered in their favor ? the army was allotted 25 percent of the seats, and the ruling party won most of the rest.

Despite fears that Suu Kyi risks legitimizing a regime she has opposed for decades, her backers see the poll as a chance to take advantage of a government-orchestrated political opening that could eventually spawn real democracy.

Suu Kyi herself said Friday that campaigning had been marred by so many irregularities that it could not considered fair ? allegations her party reiterated Sunday.

Malgorzata Wasilewska, head of the European Union's observer team, called the voting process "convincing enough" but stopped short of declaring it credible yet. "In the polling stations that I visited ... I saw plenty of good practice and good will which is very important," she said.

U.S. Rep. Joseph Crowley, who spearheaded legislation that tightened sanctions in 2008, praised Suu Kyi and the opposition for taking part in the vote, but said more needed to be done.

"Now is not the time for the international community to rush toward lifting pressure on Burma," said Crowley, who in January became the first House member to visit Myanmar in 12 years. "Far too many political prisoners are still locked behind bars, violence continues against ethnic minorities and the military dominates not only the composition but the structure of the government."

Despite the polling problems, Suu Kyi had no regrets and stayed in the race anyway.

"She's fully aware of the risks, even of the possibility that the Burmese government is attempting to co-opt her," said Sean Turnell, a Myanmar expert at Macquarie University in Sydney, Australia. But "I think she sees an opportunity, and is pushing hard to make it real."

At the very least, her candidacy has galvanized Myanmar's downtrodden masses, giving hope where only the smallest slivers existed before.

Party executive Soe Win said Suu Kyi had won in 128 of the 129 voting stations in Kawhmu, the constituency south of Yangon that she contested. He said the party had received reports of irregularities in the sole holdout.

"She may not be able to do anything at this stage," said Go Khehtay, who cast his ballot for Suu Kyi at Wah Thin Kha, one of the dirt-poor villages in the rural constituency that she is vying to represent. "But one day, I believe she'll be able to bring real change."

David Scott Mathieson, an expert on Myanmar for Human Rights Watch, said "the real danger of the by-elections is the overblown expectations many in the West have cast on them."

"The hard work really does start afterward," he said. "Constitutional reform, legal reform, tackling systemic corruption, sustainable economic development, continued human rights challenges ... will take many years."

One look at the village Suu Kyi awoke in Sunday ? where some voters walked barefoot into a schoolhouse-turned-polling station ? illustrates the long road ahead.

With no running water, women in Wah Thin Kha draw water from wells with plastic buckets attached to bamboo sticks. With no electricity, her supporters rigged up an electrical grid fueled by groaning generators to light her arrival. There is no Internet, and aside from radios and cellphones, most everyone is cut off from the modern world.

Residents said the ruling party recently began building a narrow concrete pathway through the village in an apparent last-ditch attempt to win votes. But the concrete has already begun to crack, and few appeared impressed.

"The government built a clinic here a long time ago, but we've never seen a doctor inside it," said Nini Aung, a Suu Kyi supporter whose cheeks were smeared with a decorative cream-colored paste made from ground tree bark.

"We need hospitals and clinics. We need change in months, not years," she said. "The junta never did much here. We have relied on ourselves, as if we were on our own."

Sawhkin Zaw, another voter in Wah Thin Kha, said he didn't expect anything to change soon. But he cast his ballot for her because "she's sacrificed a lot to get to this point. We need to give a little back."
